目的:研究、探讨多向药耐药基因(MDR1)及其表达产物P-糖蛋白(P-gp)、拓扑异构酶Ⅱ(TopoⅡ)、谷胱甘肽-S-转移酶(GST-л)和胸苷酸合成酶(Ts)的表达及其临床意义.方法:采用SP法对130例随访10年以上的结、直肠患者进行了癌组织中P-gp、GST-л、Topo Ⅱ和TS的表达,并分析与临床预后的关系.结果:GST-л的表达与UICC分期有关(P=0.022),而与组织类型、浸润深度及淋巴结转移无关:Pgp、TopoⅡ和TS的表达与上述参数均无关(P＞0.05).经多因素Cox比例风险模型分析显示,UICC分期、组织类型及GST-Ⅱ、TopoⅡ的表达与结直肠癌患者术后生存率有关(P＜0.05).结论:检测结直肠癌中P-gp、GST-x、TopoⅡ和TS的表达,对肿瘤化疗药物的选择具有重要临床意义;UICC分期、组织类型及GST-Ⅱ、TopoⅡ的表达似可作为结直肠癌患者独立的预后因素.
