补肾生血方剂对苯诱发再生障碍性贫血小鼠造血及免疫功能调节的研究

摘要:

目的:通过观察补肾生血方剂对苯诱发再生障碍性贫血模型小鼠免疫及造血功能的影响,初步探讨中药的调节机制。方法:将清洁级Balb/c小鼠60只随机分为正常对照组、再障模型组和补肾生血方剂治疗组3组各20只;对照组注射玉米油4ml/kg;模型组和治疗组注射苯油混合液(苯剂量为1ml/kg体重),连注14天;然后治疗组小鼠按临床等效剂量的5倍灌胃给以相当于生药量为5.92g/(kg·d)的补肾生血方药,连续4周。结果:(1)模型组和治疗组小鼠体重及脾指数和胸腺指数较对照组均下降,差异有统计学意义(P<0.01);而治疗组小鼠胸腺指数和脾指数明显高于模型组,差异有统计学意义(P<0.01)。(2)模型组和治疗组小鼠外周血WBC、RBC、Hb、PLT的水平较对照组均明显下降,差异有统计学意义(P<0.01),且以模型组最明显;而治疗组与模型组比较,除RBC外,余3项指标均高,差异有统计学意义(P<0.01)。(3)模型组和治疗组骨髓组织较对照组均有不同程度的病理改变,且以模型组最明显,而治疗组的病理变化较模型组轻微。(4)模型组和治疗组小鼠血清sIL-2含量较对照组明显升高,差异有统计学意义(P<0.01);而TNF-α含量略升高,但差异无统计学意义(P>0.05)。治疗组sIL-2含量低于模型组,差异有统计学意义(P<0.01);但TNF-α含量较模型组无统计学差异(P>0.05)。结论:补肾生血方剂能明显改善苯诱发再障小鼠模型的骨髓造血功能,并可有效调节血清TNF-α、sIL-2的水平,部分纠正了模型小鼠免疫功能的紊乱。

Objective:To observe the effects on immune and hematopoietic function in benzene induced aplastic anemia model mice by feeding tonifying kidney and generating blood prescription and preliminary study the adjustment mechanism of Chinese medicine.Methods:60 Clean-class Balb/c mice,were randomly divided into 3 groups,namely normal control group,AA model group,TKGB prescription treatment group.The normal control group was injected with corn oil,4 ml/kg;AA model group and treatment group was injected with the oil mixture of benzene(1 ml/kg in body weight),the injection lasting for 14 days.According to 5 times the clinical equivalent dose,the treatment group mice were fed with TKGB prescription(raw herbs dose 5.92g/kg d) for 4 weeks.Results:(1)Compared to control group,body weight,spleen index and thymus index in model group and treatment group were decreased,the differences were statistically significant(P<0.01);in treatment group and model group,those were increased.(2)Compared to normal group,the numbers of blood cells in model group and treatment groups were significantly decreased,the difference was statistically significant(P<0.01);the most obvious was found in model group,and the numbers of blood cells in treatment group was slightly more than model group,in addition to red blood cells,the differences were significantly different(P<0.01).(3)Compared to normal control group,there were some pathological changes of different degrees in bone marrow of model group and treatment group,the most significant pathological changes appeared in model group,while slight changes in treatment group.(4)Compared to the normal group,sIL-2 levels were significantly elevated in model group and treatment group,the difference was statistically significant(P<0.01);while the TNF-α level was increased slightly,but the difference was not statistically significance (P> 0.05);sIL-2 level in treatment group was lower than the model group,the difference was statistically significant(P<0.01);TNF-α level was slightly higher than the model group,the difference was not statistically significant(P>0.05).Conclusions:From point view of tonifying kidney,the tonifying kidney and generating blood prescription could significantly improve bone marrow hematopoietic function of benzene-induced aplastic anemia model mice of,to a certain extent,effectively repaired the bone marrow pathological damage caused by benzene,meanwhile effectively control the serum TNF -α,sIL-2 levels by regulating model mice immune function.
