摘要:目的探讨中药克疣灵对尖锐湿疣(CA)病人NK细胞凋亡调控蛋白sFas及其表面活性因子sCD226表达水平的影响作用,阐明克疣灵的正向免疫调节作用。方法对80例男性CA病人随机分为克疣灵高、低剂量〔量取1 ml加入无菌去离子水稀释成100 mg/ml(高剂量)、25 mg/ml(低剂量)备用〕组、IL\|15组、空白对照组,每组各20例,25例健康男性为正常对照组,抽取受试者静脉血,采用自然沉降法获取白细胞、Ficoll-Urog分层液离心处理获取非黏性淋巴细胞、Percoll非连续梯度密度离心处理获取NK细胞,并进行NK细胞与不同剂量的克疣灵体外共同培养,采用ELISA法于培养48、96 h 测定NK细胞凋亡调控蛋白sFas及其表面活性因子sCD226的表达水平。结果克疣灵高剂量组在培养48 h 后NK细胞凋亡调控蛋白sFas表达水平显著下调,其表面活性因子sCD226表达水平显著提高,上述指标与IL-15组比较差异无统计学意义(P>0.05),与克疣灵低剂量组、空白对照组比较差异有统计学意义(P<0.05或<0.01)。结论高剂量克疣灵能显著降低NK细胞对凋亡的易感性,提高其细胞活性,具有较强的正向免疫调节作用。
Abstract: Objective To clarify the positive adjustment effect by the experimental study of the effect of keyouling on expression of sFas and CD226 in the NK cell of the patients with condyloma acuminata(CA).Methods After isolated the NK cell from the peripheral blood of 80 male CA patients by Ficoll-Hypaque centrifugation, the NK cell was cultured with different dosage of keyouling.To determine the expression of sFas and CD226 in the NK cell by means of ELISA after cultured 48 and 96 hours.Results The expression of sFas in NK cell l had been significantly reduced by the high dosage of keyouling after cultured 48 hours.The expression of CD226 in NK cell l had been significantly reinforced by the high dosage of keyouling after cultured 48 hours.There were no significant difference of all the above detail compared with the group of IL-15(P<0.05),but there were significant difference compared with the groups of the low dosage of Keyoulingand and blank control group(P<0.05,P<0.01).Conclusion The high dosage of keyouling could reduced the abnormal apoptosis of the NK cell of the patients with CA.It had significant positive immune adjustment effect.
